In your agenda custom command first block:

  ...
  (org-agenda-tag-filter ("+Tag"))
  ...

In your agenda custom command second block:

  ...
  (org-agenda-tag-filter ("-Tag"))
  ...

Not tested, but it should work -- not the -/+ before the tag itself,
that's what conditions tag filtering out/in.
