Perhaps it should be the sole entry, perhaps you have other org files somewhere else that you want to include in the agenda; but if the directory is in the list, then the agenda will pick up items from newly created files in that directory, e.g. with org-agenda-redo (that's bound to "g" in the agenda buffer).
